Biosensors for the detection and monitoring of Alexandrium minutum along the French coasts ArchiMer

The microalgae Alexandrium minutum produces neurotoxins responsible for paralytic shellfish poisoning, which upon accumulation in shellfish represent a human health risk. This species recurrently forms toxic blooms in the Northern part of the French Brittany coast. At present, algae identification relies on tedious microscope observations mostly performed by skilled taxonomists in laboratories. Newly emerging fields of molecular taxonomy, nanoscale technology and biotechnology offer opportunities for the development of in-situ biosensors for the detection and the monitoring of toxic algae.

Detection and molecular identification of Pseudo-nitzschia species in natural samples from the French coasts ArchiMer

Domoic acid (DA) was first detected in France in mussel flesh in 1998, outbreak that was linked to the presence of Pseudo-nitzschia pseudodelicatissima. Ever since, the phytoplankton monitoring program (REPHY, Réseau de surveillance du Phytoplancton et des Phycotoxines) also monitors DA content in shellfish flesh. Amnesic shellfish poisoning (ASP) has been recurrently detected, and was over the sanitary threshold in the years 2000 and 2004 in Brittany and in 2002 in the Mediterranean coast (Amzil, 2001; Amzil pers. comm.). Direct detection of toxic micro-algae Alexandrium minutum through high sensitive gravimetric sensors ArchiMer

In this work QCM technologies were used to set up a DNA based biosensor dedicated to the detection of toxic micro-algae in the environment. Up to now, appropriate detection were not achieved for different reasons as the possibility of regeneration, the capability of multidetection or the detection of very low DNA target concentration. For these reasons different configurations were examined based on in-situ and fast regeneration, supramolecular chemistry of DNA and high sensitive gravimetric devices. In late 2004, the French phytoplankton monitoring network (REPHY of Ifremer) detected domoic acid (DA) above the EU-regulatory limit of 20 μg DA/g tissue in king scallops (Pecten maximus) from the Bay of Seine in the Channel (Fig. 1). In situ QCM DNA-biosensor probe modification ArchiMer

We report on a probe modification of a quartz crystal microbalance (QCM) DNA-biosensor that permits to reversibly change the DNA sequence detected. A QCM DNA-biosensor was designed by immobilization of a 20-base DNA-disulfide probe on the gold-covered quartz surface of a 27 MHz microbalance (9 MHz, third overtone). After immobilization on the gold covered quartz surface, this probe was modified by hybridization with a 45-base DNA that includes the complementary 20-base sequence, a 5-base spacer and a non-complementary 20-base sequence. The non-complementary sequence constitutes a new probe, different from the DNA-disulfide probe, that permits the detection of a new DNA target. Interaction protection cathodique et biofilm en milieu marin ArchiMer

In marine environments cathodic protection using sacrificial anodes or impressed current is widely used. In aerated seawater a cathodic potential of – 800 mV/SCE is generally accepted as efficient against marine corrosion. Biofilms formed on the metallic surfaces exposed to seawater can promote localized corrosion and interfere with the cathodic polarization process. Experiments were conducted both in natural environments and with a monoculture of a marine bacterium in order to understand the relationship between cathodic protection and biofilm formation. ISSR as new markers for genetic characterization and evaluation of relationships among phytoplankton ArchiMer

In order to increase the molecular tools and markers needed for the identification of phytoplankton species, the inter simple sequence repeat ( ISSR) fingerprinting was adapted to micro-algae and its use in genetic analysis was demonstrated. Twelve strains, 6 Alexandrium, 4 Pseudo-nitzschia, 1 Skeletonema and 1 Tetraselmis were analysed for the first time with ISSR amplifications. The patterns were highly polymorphic and very reproducible. The 6 primers gave 223 polymorphic markers that clearly and easily distinguished all 12 strains ( mainly toxic ones) and gave 187 polymorphic markers among the Alexandrium and the Pseudo-nitzschia species. Layer-by-Layer DNA film synthesis via branched hybridization ArchiMer

A new Layer-by-Layer (LbL) DNA film synthesis, where the driving force is the natural DNA hybridization, is presented in this work. The DNA films were synthesized via a branched mechanism and do not include any chemical binders between each layer as it is the case for film obtained by other synthesis paths. Kinetics of film formation were monitored by mass measurements with an electroacoustic network analyzer. The presented synthesis is a pathway to new DNA structures which can be used in biotechnologies and is complementary to those obtained by other LbL techniques.

Oligonucleotide quartz crystal microbalance sensor for the microalgae Alexandrium minutum (Dinophyceae) ArchiMer

Toward Detection of Harmful Algae Blooms by in situ Surface Plasmon Resonance Spectroscopy ArchiMer

Among marine algae species, Alexandrium minutum produces a phycotoxin called paralytic shellfish poisoning (PSP) that is introduced in the food chain through the ingestion of phytoplankton by shellfishs, and later by human consumers. Thus, in situ monitoring of A. minutum proliferation in coastal seawater is of great economical importance for marine resources exploitation. Here, we propose a rapid test for the detection of A. minutum by surface plasmon resonance spectroscopy. First, whole genomic DNA is extracted from the algae. Second, a 677 bp long portion of the 28S ribosomal DNA is amplified by PCR. Third, the PCR product is detected by surface plasmon resonance spectroscopy onto a DNA functionalized gold substrate.

Toward in situ detection of algae species ArchiMer

For the last few years there is a more and more pressing need to develop system for detecting HAB at their early stage. Researchers have then been working on new in situ sensor. In this paper a submarine transducer based on surface plasmon resonance is reported. It demonstrated a refractive index resolution of 3.10(-6), which is promising for later phytoplankton detection.
